Steven Gerrard has confirmed (official Twitter) that the first-team duo are doubts before the Sunday match against Aberdeen at Ibrox.
Gers captain Tavernier has a knee problem, while veteran centre-forward Defoe has a tight muscle.
The Light Blues have just four games remaining in the season and are second in the league table, with an eight-point cushion over third-placed Aberdeen.
A victory at the weekend will make sure of a second-placed finish, but they may have to do it without some key players.
Alfredo Morelos is still serving a four-game suspension and they could be without another key striker in Defoe.
Defoe, 36, has scored seven goals and supplied seven assists in just 16 games since he linked up with the Scottish giants in January on loan.
